Cables definitely matter, both signal and speaker. Anybody who says otherwise either has faulty ears or else tested a cable already similar to was being replaced.
Voodoo cables are great, as are Mogami and even George L's. Mogami is very transparent and clear, almost hi-fi/sterile sounding. They are great for anything after the main cable from guitar. Such as connections between pedals, etc, and the final run to the amp. Same for George L's and a few other types. The cable from the guitar to the first input (whatever input that may be) is where it's worth doing some experimenting as different cables can color the tone and sometimes even feel slightly different.
Speaker cables are more straightforward, but go with something decent size (14 guage or bigger) with decent connectors. The deepness of the low end and the clarity of the overall sound can be better appreciated with the right speaker cables. More specifically, what often happens is that over time connectors begin to corrode (not always visibly) and the wiring begins to oxidize (especially with copper) so a speaker cable can eventually introduce undesired effects on the sound via reaction between speaker and output transformer, which although subtle, can definitely be audible to those humans without excess hair in their ears...unlike DC. ;D
